When should I outsource gene synthesis rather than cloning from a cDNA library?

Outsource synthesis when the target sequence is rare in available libraries. Synthesis also helps when you need codon optimisation for a non-native host. Use it too when the sequence needs changes, such as restriction site removal, tag insertion, or removal of toxic codons.

Synthesis is cost-effective for genes shorter than 3 kb. For these short genes, PCR-based cloning from genomic DNA is often unreliable.

What information do I need to provide when requesting a subcloning or gene synthesis service?

You need the target sequence in FASTA or GenBank format. You also need the destination vector name and sequence map, and the intended expression host.

Include any required tags or fusion partners, and where they should sit on the construct. Specify if you need codon optimisation, and note any preferences for restriction sites or assembly methods.

How long does Sanger sequencing take compared with next-generation sequencing for clone verification?

Sanger sequencing returns chromatograms for individual clones in one to two business days. It is the standard method to confirm a single construct before scale-up.

Next-generation sequencing works better for larger jobs. Choose it when you need to verify a pooled library, screen many variants at once, or detect low-frequency mutations in a population.

Are turnaround times and pricing fixed for all service categories?

Turnaround and pricing are on request for gene synthesis, cloning, and sequencing services, because project complexity varies. Sanger sequencing has published standard turnaround times.
