Protein detection and quantification services run on the samples you send

Three testing services sit under protein detection and quantification: array testing, ELISA testing, and Simoa testing. Partner RayBiotech manufactures over 700 ISO certified and rigorously tested multiplex arrays along with a selection of high quality ELISA kits, and those kits form the basis of the services described here.

The working pattern is the same in each case. You send your samples, the assay is run by the RayBiotech Service Department, and analysis data comes back to you. Work is carried out by a team of highly experienced scientists in a GLP-compliant laboratory, which is what holds accuracy and reproducibility steady from one batch of your samples to the next.

That arrangement helps when a platform is not installed in your own laboratory, when the analyte sits below the range of the assays you already run, or when you would rather keep bench time for the biology. Reagents for work you run yourself sit under antibody and protein arrays and ELISA kits.

Choosing the right protein detection service for your project

  1. Array testing services. Measuring many targets in one run is the reason to send samples for array testing. Full testing services are available for all RayBiotech array kits, including custom arrays and both quantitative and semi-quantitative platforms, so the format can be matched to your target list. Antibody array options run from Quantibody quantitative sandwich glass slides, through semi-quantitative sandwich arrays on membrane or glass, to semi-quantitative label-based high-density arrays, and RayPlex quantitative bead arrays read by flow cytometry.
  2. Protein arrays and glycobiology arrays. Binding questions are served by protein arrays featuring cytokines, allergens, cancer-related proteins and more, which support profiling of the serological antibodies in your samples and of protein-protein interactions. Glycobiology arrays detect glycan binding partners, glycosylation protein profiles, and glycan profiles using lectins. Both array families are manufactured by RayBiotech and run by its service department.
  3. ELISA testing services. A defined target and a quantitative number point towards ELISA for your project. The service covers sandwich, indirect, phosphorylation, competitive and cell-based formats, along with IQELISA immuno-PCR for ultrasensitive low-consumption work, transcription factor activity assays, and binding assays for inhibitor screening. Seventeen target species are covered, including rabbit, dog and zebrafish.
  4. Simoa testing service. Very low concentrations and small sample volumes are where Simoa fits your work. RayBiotech describes its Simoa testing as 1000 times more sensitive than ELISA, reaching femtogram per millilitre levels in serum, plasma, CSF and other complex matrices, with multiplexing available for up to 4 analytes.

Multiplex cytokine measurement

Quantibody glass slide arrays give quantitative readings for many cytokines in a single run, so each sample you send yields far more data points.

Expression screening before quantitation

Semi-quantitative sandwich and label-based arrays give you a broad view of protein expression, narrowing the target list ahead of quantitation.

Low abundance biomarker measurement

Simoa reaches femtogram per millilitre levels in serum, plasma and CSF, bringing analytes into range when your sample volume is limited.

Signalling readouts

Phosphorylation and cell-based ELISAs detect phosphorylated proteins in your lysates or adherent cell lines, giving a semi-quantitative view of activation.

Serological antibody profiling

Protein arrays carrying cytokines, allergens and cancer-related proteins profile the antibodies in your serum samples and map protein-protein interactions.

How does Simoa reach concentrations that ELISA cannot measure?

Simoa isolates individual immunocomplexes on paramagnetic beads using standard ELISA reagents. The difference from a conventional immunoassay is that single molecules are trapped in femtolitre-sized wells, which gives a digital readout for each bead as either bound to the target analyte or not.

Traditional ELISA is limited to picogram per millilitre detection. Simoa can achieve sensitivity as low as femtogram per millilitre levels, which brings biomarkers into range that were previously difficult or impossible to measure. RayBiotech describes the result as 1000 times more sensitive than ELISA. Simoa also requires a dedicated detector, so samples are shipped in and RayBiotech runs the assay and returns a data report.

Which ELISA formats are available through the service?

Sandwich ELISA for quantitative protein detection, indirect ELISA for detection of antibody response to pathogens, phosphorylation ELISA for semi-quantitative detection of phosphorylated proteins in lysates, and IQELISA immuno-PCR for ultrasensitive immuno-quantitative detection with low sample consumption.

Also available are competitive ELISA for quantitative competition-based detection of soluble factors, cell-based ELISA for direct detection of phosphorylated proteins in adherent cell lines, transcription factor activity assays for detection of transcription factor activation, and binding assays for competition-based inhibitor screening.

Can an array be run in house and scanned through the service?

Yes. A free slide-scanning service is offered for all glass slide antibody arrays, for customers who run their own array without access to a compatible laser scanner. Only shipping charges apply.

Who manufactures the kits used in these services?

The complete range of arrays is manufactured by RayBiotech, as are the ELISA kits used in the ELISA testing service. The portfolio can be completed further with custom arrays.
