SparQ Cumate Switch Starter Pack

Product Description

Get started with the SparQ™ Cumate Switch System, with the SparQ Cumate Switch Starter Pack. The Starter Pack includes all three components of the SparQ Cumate Switch System so you can quickly get cumate-inducible expression of your gene- or miRNA-of-interest into your lab.

Choose any inducible cloning vector listed below, and also receive CymR-T2A-Puro Lentivector and Cumate solution (1000x).

Choice of Inducible Vectors:

Indicate your inducible lentivector when placing your order.

SparQ IRES Vectors:

The IRES vector configuration enables the inducible expression of a microRNA or a cDNA along with an IRES-mediated GFP or RFP marker that will be co-expressed to monitor induction when the switch is turned ON.

pCDH-CuO-MCS-IRES-GFP co-inducible IRES lentivector
pCDH-CuO-MCS-IRES-RFP co-inducible IRES lentivector

SparQ T2A Vectors:

The T2A vector configuration enables the inducible expression of a cDNA along with the simultaneous, T2A-mediated GFP or RFP marker to monitor cumate switch induction.

pCDH-CuO-MCS-T2A-GFP co-inducible T2A lentivector
pCDH-CuO-MCS-T2A-RFP co-inducible T2A lentivector

SparQ Dual Promoter Vectors:

The dual promoter vector configuration enables the inducible expression of a microRNA or a cDNA. The downstream EF1 promoter will constitutively express RFP or GFP or RFP/GFP plus puromycin independent of the cumate switch activation.

pCDH-CuO-MCS-EF1-GFP dual promoter lentivector
pCDH-CuO-MCS-EF1-RFP dual promoter lentivector
pCDH-CuO-MCS-EF1-GFP+Puro dual promoter and dual marker lentivector
pCDH-CuO-MCS-EF1-RFP+Puro dual promoter and dual marker lentivector

How it works

The regulatory mechanisms of the bacterial operons cmt and cym have been engineered to regulate gene expression in mammalian cells. In the repressor configuration, regulation is mediated by strong binding of the CymR repressor to the Cumate operator site (CuO), which is downstream of the CMV5 promoter. Addition of cumate (depicted as yellow circle in the graphic scheme below), a non-toxic small molecule inducer, relieves the repression and drives gene activation; in this case, fluorescent proteins (RFP and GFP). Subsequent removal of cumate from the growth media reverts the activation process, rendering gene switches to off mode.
