Product Description

Designed to transiently deliver PhiC31 Integrase for your PhiC31 and PinPoint genome engineering studies, the PhiC31 Integrase Expression Plasmid is an essential component of these one- and two-step non-viral gene delivery systems. PhiC31 integrase can integrate a plasmid of any size, as a single copy, and requires no cofactors. The integrated transgenes are stably expressed and heritable.

The phiC31 Integrase System can also be ulitized as a nonviral technology for transgenesis. With both iPSC-generating donor and PhiC31 Integrase Expression plasmids, the Non-Viral PhiC31 Integrase Mouse iPSC Reprogramming Kit delivers a complete, non-viral integrase-based iPSC generation system. Figure 1. The PhiC31 Integrase System efficiently introduces a puromycin resistance marker to HEK293 cells. The dual promoter PhiC31 Donor Vector pFC-PGK-MCS-EF1-GFP-Puro was co-transfected into HEK293 cells with either carrier DNA (top plate) or the PhiC31 Integrase Expression Plasmid (bottom plate) and the cells placed under puromycin selection after three days. After an additional eleven days, the cells were fixed and stained with a solution of 50% methanol plus 1% methylene blue to image colonies. Only the cells co-transfected with integrase formed colonies (bottom panel), demonstrating the high efficiency of the PhiC31 Integrase System for transgene delivery.
