One-step probe-based RT-qPCR master mixes combine reverse transcription and TaqMan-probe qPCR in a single tube. - Enzyme blend. The RT and DNA polymerase must work in compatible buffers. Some blends limit one-step performance compared to two-step.
- Sensitivity at low copy. For very low-copy viral load two-step often outperforms one-step.
- Multiplex capability. Validated for 4- to 6-plex on standard cyclers.
- Workflow time. One-step adds an RT step, lengthening cycle time but eliminating a handling step.
