One-step SYBR-based RT-qPCR mixes for cost-effective high-throughput screening

One-step SYBR-based or intercalating-dye RT-qPCR mixes combine reverse transcription and qPCR in a single tube with SYBR detection. The format is the cheapest one-step option, useful for high-throughput screening, assay development, or applications where probe-based specificity is not required.

Pick by chemistry and sample input.

  1. Chemistry. SYBR or EvaGreen; both work with standard hot-start chemistries.
  2. Inhibitor tolerance. Some formulations handle direct biofluid or crude lysate input. Useful for high-throughput screening.
  3. Specificity verification. Always run a melt-curve in initial validation. SYBR has no inherent specificity beyond primer design.
  4. Sensitivity. Slightly lower than two-step for low-copy targets.
  5. Cost per reaction. Lower than probe-based; useful for high-volume routine work.
