The Gibson Assembly Ultra method utilizes a special mix of enzymes to assemble DNA quickly and efficiently in a two-step reaction. Multiple DNA fragments can be joined with 95% cloning efficiency and constructs up to 1000 kb in length can be efficiently assembled by designing multi-stage Gibson Assembly Ultra reactions.

How It Works

The Gibson Assembly Ultra method utilizes a special mix of enzymes to assemble DNA quickly and efficiently in a two-step reaction. Multiple DNA fragments can be joined with 95% cloning efficiency. Constructs up to 1000 kb in length can be efficiently assembled by designing multi-stage Gibson Assembly Ultra reactions.

  1. Design DNA fragments with overlap regions
  2. Combine the fragments and Ultra Master Mix A => Master Mix A catalyzes 3’ chew back
  3. Inactivate Ultra Master Mix A at 75°C
  4. Anneal DNA fragments at 60°C
  5. Combine the fragments and Ultra Master Mix B >> Master Mix B catalyzes repair

The resulting construct is double stranded, fully sealed, and can be used immediately for downstream molecular biology applications
