1. Cost-sensitive routine. Basic Taq for colony screening, genotyping.
2. Higher specificity. Hot-start Taq for reduced primer-dimer.
3. Convenience. 2X master mix for fewest pipetting steps.
4. Gel loading. Dye-loaded mix for direct gel loading.
5. Inhibitor tolerance. Direct-PCR formulations for crude lysate (separate category).
6. Fidelity caveat. Basic Taq error rate is ~10^-5 per base. Fine for genotyping but not for cloning long products. For cloning use high-fidelity (separate category).
