- Cross-sample normalisation. Use reference RNA pooled from multiple tissue types. Acts as inter-experiment calibrator.
- Input-control spike-in. Use synthetic spike-ins (ERCC for RNA-seq, ddPCR-validated DNA spike-ins for qPCR) to monitor pipeline performance.
- Standard curve template. Use templates of defined copy number for absolute quantification.
- Certification level. For absolute quantification copy-number standards must come from a defined source with batch records; without that, claims of absolute concentration are not defensible.
