Probe-based qPCR master mixes for high-specificity multiplex and allelic discrimination assays
TaqMan-style hydrolysis-probe qPCR master mixes give higher specificity than SYBR-based detection because amplification signal requires both correct primer binding and correct probe hybridisation. The format is essential for multiplex qPCR (different fluorophore-quencher pairs per target), allelic discrimination, and clinical-research applications where false positives matter.
Pick chemistry and instrument by needs.
- Chemistry. Standard hydrolysis-probe is the workhorse. For higher specificity allelic discrimination use Molecular Beacon dual-probe formats.
- Instrument compatibility. ROX reference dye level matters for Applied Biosystems instruments; passive-reference-free for some newer cyclers. Check before ordering.
- Multiplex capability. Validated for 4- to 6-plex on standard cyclers. For multiplex use mixes specifically validated for multiplex; some single-target mixes lose sensitivity beyond duplex.
- Sensitivity. For low-copy targets choose mixes with documented detection down to single-copy.
- Inhibitor tolerance. For crude lysate input choose mixes formulated to tolerate inhibitors.
