Isothermal amplification chemistries for nucleic acid detection without thermocycling

When you need to amplify nucleic acid at a single temperature without thermocycling, isothermal chemistries (LAMP, RPA, NASBA, HDA, SDA) do it. Useful for point-of-care diagnostics, field deployment, and applications where thermocycler access is limited or rapid results matter.

Pick chemistry by deployment and target.

  1. Chemistry. LAMP highest sensitivity and most published. RPA fastest and lowest temperature. HDA simpler primer design. NASBA RNA-specific.
  2. Reaction format. Lyophilised for room-temperature shipping. Liquid for higher sensitivity.
  3. Detection. Turbidity for naked-eye. Fluorescence for quantification. Colorimetric pH-shift for point-of-care.
  4. Sensitivity. LAMP can detect 10 copies. RPA detects similarly with shorter time but more primer-design constraints.
  5. Regulatory. For point-of-care diagnostic development LAMP has the most regulatory precedent.
