Exosome engineering tools for cargo loading, surface display, and EV tracking

Loading cargo into an exosome without damaging the vesicle is the central challenge in EV engineering, and this range of more than 90 products lets you match the approach to your experiment. Lentiviral vectors express EV-enriched scaffold fusion proteins to package heterologous protein cargo into secreted EVs, while dedicated XMIR and XMIRXpress vectors load specific (anti)miRNA sequences without electroporation or sonication. Surface display constructs present targeting peptides or full-length proteins on the exosome surface using scaffolds such as the C1C2 domain of MFG-E8 (XStamp tag). The range also includes EV-Luminite NLuc bioluminescent reporters and related stable cell lines, working with stably transduced producer lines or transient transfection.

Pick by cargo and targeting.

  1. Protein loading. Choose XPack-based fusion systems. Cargo packaged into the exosome lumen during biogenesis.
  2. RNA loading. Use RNA-binding-protein fusion to exosome-membrane proteins. Different chemistry to protein loading.
  3. Surface targeting. For generic membrane display use CD63 fusion. For specific tropism use engineered tetraspanins.
  4. Production host. HEK293T is standard for research. Mesenchymal stem cells for immune-privileged therapeutic candidates.
  5. Tracking. Use luciferase or fluorescent fusions for in vivo tracking; verify tags do not alter biogenesis kinetics.
