SYBR Green and EvaGreen dye-based qPCR master mixes for assay development
SYBR Green and EvaGreen intercalating-dye master mixes give cheaper qPCR than probe-based, useful for assay development, melt-curve analysis, and applications where primer-design specificity is sufficient. The choice between dye and probe depends on specificity needs.
Pick by chemistry and specificity needs.
- Dye choice. SYBR Green is standard. EvaGreen has lower polymerase inhibition; useful for some difficult templates.
- Hot-start. Essential for specificity. Aptamer-based and antibody-mediated hot-start give cleaner amplicons than wax-based.
- Reference dye. Match ROX or passive-reference-free format to your instrument.
- Verification. For new assays always run a melt-curve and gel confirmation in early validation. SYBR specificity depends entirely on primer design.
- Multiplex caveat. SYBR cannot multiplex (single dye). For multiplex switch to probe-based.
