Arima-HiC+ FFPE Kit
The Arima-HiC+ FFPE Kit and workflow are ideal for the detection and discovery of structural variants from archival or clinical samples
Product Description
Hi-C is a proximity ligation method that captures the three-dimensional organizational structure and long-range gene regulatory interactions of chromatin. Detect structural variants in the genome using the Arima-HiC+ FFPE kit. With its robust and easy-to-use workflow, your formalin fixed paraffin embedded (FFPE) samples can yield critical genomic information — regardless of sample age.
- Proven Performance - Use Arima’s proven HiC+ technology to generate high-quality data from clinically relevant samples
- Dependable Results - Generate data you can trust, with built-in QC steps to ensure you get reliable sequencing results every time
- Increased Discovery Power - Capture more data from FFPE samples to detect structural variants and their impacts on gene regulation
- Detect both coding and non-coding structural variants
- Link structural variants to impacts on gene function
- Efficiently generate multiple data types from a single assay
The Arima-HiC+ FFPE kit provides dependable results from this hard-to-use sample type that are highly correlated with results from fresh-frozen tissue samples.
Arima-HiC+ FFPE Workflow
With Arimas easy-to-use workflow you can rapidly go from sample to discovery. Arima Hi-C is an experimental workflow that capture a genome’s sequence and structure (three-dimensional conformation). During this workflow the FFPE tissue is crosslinked with formalin prior to embedding into Paraffin way. Thin sections are made from the tissue using a microtome and the sections are dewaxed and rehydrated then the cross-linked chromatin is digested using a restriction enzyme (RE) cocktail. The 5’-overhangs are then filled in, causing the digested ends to be labeled with a biotinylated nucleotide. Next, spatially proximal digested ends of DNA are ligated, capturing the sequence and structure of the genome. The ligated DNA is then purified, producing pure proximally-ligated DNA. The proximally-ligated DNA is then fragmented, and the biotinylated fragments are enriched. The enriched fragments are then subjected to a custom library preparation protocol utilizing the 2STM Plus DNA Library Kit from IDT technologies.
