Most colorimetric and fluorometric kits accept cell lysates, tissue homogenates, cell culture media, blood plasma, and urine. Lysis buffer composition matters: detergent-based buffers can interfere with some enzyme activity assays. We recommend that you normalise protein concentration with a BCA or Bradford assay when you compare across sample types.
Use mitochondria-targeted fluorescent probes to selectively detect mitochondrial superoxide. These probes build up in the mitochondrial matrix because of membrane potential. Measure cytoplasmic ROS with membrane-permeable dyes instead. These dyes are oxidised by a broad range of reactive species throughout the cell.
