  1. Cargo. Match reagent chemistry to cargo class (DNA, siRNA, mRNA, RNP). Some reagents work for multiple cargo classes; others are cargo-specific.
  2. Cell type. HEK293 and HeLa transfect with almost anything. Primary T cells, neurons, and pluripotent stem cells need specialised reagents or electroporation.
  3. Serum compatibility. Serum-free transfection medium often required during the transfection step.
