Stellaris DesignReady probe sets for single-molecule mRNA detection in fixed samples
When you need single-molecule mRNA detection in fixed cells or tissues, and your target is in the catalogue, DesignReady Stellaris probe sets give you each transcript as a punctate dot you can count. Each set is 25 to 48 short oligos labelled with the same fluorophore that hybridise along your target mRNA.
Work the decision.
- Catalogue match. If your transcript is in the DesignReady catalogue, order it. If not, switch to Customised probe sets.
- Dye and filter set. For Cy3 channel choose Quasar 570. For Cy5 channel choose Quasar 670 (also for tissues with strong green-channel autofluorescence). For three-colour multiplex add CAL Fluor Red 590 or CAL Fluor Orange 560.
- Sample type. Cultured cells and fresh-frozen tissue work with the standard protocol. For paraffin-embedded tissue use the FFPE protocol.
- Tissue thickness. Thin sections under 20 microns work with the standard protocol. For thicker tissue pilot signal depth before committing.
- Multiplex plans. Plan dye combinations against your microscope filter set in advance; spectral overlap is the most common multiplex failure. LGC Biosearch carries the catalogue.
