- Transposase delivery. Plasmid co-transfection versus mRNA versus stable transposase cell line. mRNA gives transient expression with cleanest excision.
- Selection compatibility. Match marker to your selection scheme.
- Verification. For single-copy integration use ddPCR or Southern. PiggyBac integrates randomly; verify integration count and site for quantitative work.
