Pick by target site and payload.
- attP target site. Genomic pseudo-attP sites are characterised. Some are higher-expressing or more stable than others.
- Payload size. PhiC31 accepts large constructs without efficiency penalty.
- Integrase delivery. Transient plasmid expression versus mRNA versus protein. Protein delivery gives transient expression with minimum genomic footprint.
- Single-copy verification. Verify by ddPCR or Southern.
- Use case. PhiC31 has fewer landing pads than engineered safe-harbour systems. Choose based on expression-consistency requirements.
