CRISPR/Cas9 sgRNA Libraries

Pooled CRISPR/Cas9 sgRNA libraries for genome-wide loss-of-function and gain-of-function screens

Pooled sgRNA libraries enable genome-wide or focused loss-of-function, gain-of-function (CRISPRa), or repression (CRISPRi) screens in mammalian cells. Libraries are lentiviral with 3 to 6 guides per gene, supplied as plasmid pools or pre-packaged virus, with validated benchmark genes (essential and non-targeting controls).

Pick by scope and modality.

  1. Library scope. Genome-wide for unbiased discovery. Focused (kinase, druggable, lncRNA, epigenetic) when you have a tighter hypothesis.
  2. Screen modality. Knockout (Cas9 plus sgRNA). CRISPRi (dCas9-KRAB plus sgRNA). CRISPRa (dCas9-VPR plus sgRNA).
  3. Guide design generation. Newer libraries with optimised seed-region rules outperform first-generation libraries by 2- to 3-fold in hit-rate.
  4. Species. Human and mouse common. Rat available. Other species less mature.
  5. Format. Plasmid pool versus ready-to-use virus. One-vector all-in-one versus two-vector requiring Cas9 cell line.
  6. Coverage. For positive-selection screens 500 to 1,000 cells per guide. For negative-selection drop-out screens at least 200 cells per guide.
